Mesenchymal Stem cells (MSCs) are essential players in the process of muscle regeneration. However the mechanisms involved are not well understood. Recently, our group demonstrated in the mouse model that MSCs from the subcutaneous adipose tissue (ScAT) migrate to the muscle following muscle injury in order to promote regeneration. To better understand the underpinning mechanisms, we conducted experiments using tagged ScAT engrafted in wild-type mice. We then isolated MSCs and performed single-cell RNA-seq (scRNA-seq). Importantly, we identified 2 distinct sub-population of MSCs that are recruited in two distinct conditions but all display features of regenerative MSCs that we will call "super-MSCs". We have also identified a common marker of these populations. This research will give invaluable insights into the mechanisms underlying muscle regeneration which is impaired during aging and is a cause for muscle atrophy and decreased mobility of aged individuals. These studies will be the basis for future work on MSCs and their use in cell therapy.
