The emergence of high-throughput technologies and large-scale biobanks has reshaped analytical strategies in modern human genetics. In this seminar, I will outline key questions that human geneticists aim to address and demonstrate how diverse omics datasets can be leveraged to investigate an understudied trait, premature hair graying. Hair graying is one of the most visible markers of human aging, and premature hair graying refers to its onset earlier than the typical age. Although hair characteristics are known to be highly heritable, the genetic basis of premature hair graying remains largely unexplored. In this study, we conducted a genome-wide association analysis of premature hair graying in 2,539 self-reported cases and 19,676 controls of Han Chinese ancestry, identifying thirteen associated loci. We then integrated multi-omic datasets from human hair follicles and skin tissue to highlight functionally relevant genes near these loci. Furthermore, phenome-wide association analyses suggest shared genetic mechanisms between premature hair graying and both skin and breast cancers. Our findings elucidate the genetic regulation underlying premature hair graying and offer new insights into the biology of hair aging.
