Molecular genetic approach using DNA recombinases, such as Cre and Flp, and nucleases, Cas9, have been applied to address various biological hypotheses. These genetic technologies enable for manipulating gene expression and engineer genome precisely to uncover the molecular mechanism underlying a variety of biological activities. However, precise spatiotemporal control is still limited though using the existing tools. In this seminar, I will summarize the history in the field of tool development and our ongoing research to develop a new set of genetic tools with single cell resolution using blue and red lights to address this limitation. In addition, I will share our progress on development of new genetically encoded fluorescent indicators for metabolites. Our proposed novel technology will be widely applicable to various research fields to address a variety of biological questions.
